Those messages travel upward to the brainstem’s nucleus tractus solitarius (NTS), the main hub where the brain processes information about internal balance.

Once the brain interprets that inflammation is underway, it responds by sending signals back down the vagus nerve’s motor (efferent) fibers.

This begins what Tracey called the inflammatory reflex, the body’s built-in mechanism to keep inflammation in check. .

Through this pathway, the vagus nerve releases acetylcholine, its primary neurotransmitter, which binds to specialized α7 nicotinic acetylcholine receptors (α7nAChR) located on immune cells such as macrophages. When these receptors are activated, they instruct the immune cells to reduce the production of pro-inflammatory cytokines like TNF-α, IL-1β, and IL-6.

In simple terms, the vagus nerve “responds with calm” by sending biochemical instructions that quiet inflammation before it spirals out of control.
